"FLOTUS" is a portmanteau of "POTUS" and "flirt", because in the early US, the president was only allowed to flirt with his wife.

The reason you see the modern word "POTUS" written in all caps is that Latin didn't have lowercase letters.

"POTUS" comes from the Latin "potus", meaning "the act of drinking alcohol", believed to have originated from the heroic amounts of alcohol consumed by the US founders after the signing of the Declaration of Independence.
